目的:观察变应性接触性皮炎Treg/Th17细胞平衡的变化,探讨龙胆泻肝汤治疗变应性接触性皮炎的部分干预机制。方法:将40只小白鼠随机分为4组:空白组、模型组、龙胆泻肝汤组、氢化可的松琥珀酸钠组,每组10只。采用DNCB皮肤涂抹方法制备小鼠背部ACD模型。模型复制成功后,龙胆泻肝汤组、氢化可的松琥珀酸钠组小鼠均按照成人2倍等效剂量灌胃,连续给药21 d,空白组、模型组给予等量蒸馏水灌胃。实验结束后,取小鼠致敏部位皮肤,对皮肤组织中Foxp3、RORγt、IL-17 mRNA及蛋白的表达水平进行检测。结果:与空白对照组比较,模型对照组、西药组、龙胆泻肝汤组小鼠皮肤组织中Foxp3 mRNA及蛋白表达水平显著降低(P<0.01);而IL-17、RORγt mRNA及蛋白表达水平显著升高(P<0.01)。与模型对照组比较,西药组和龙胆泻肝汤组小鼠皮肤组织中Foxp3 mRNA及蛋白表达水平显著升高(P<0.01);而IL-17、RORγt mRNA及蛋白表达水平显著降低(P<0.01)。结论:Treg/Th17细胞失衡可能是变应性接触性皮炎的发病机制之一,而龙胆泻肝汤可能就是通过作用于该靶点,调节Treg/Th17细胞平衡,而实现其对变应性接触性皮炎的治疗作用。
Objective: To observe the changes of balance of Treg / Th17 cells in patients with allergic contact dermatitis and to explore some intervention mechanisms of Longdan Xiegan Decoction in the treatment of allergic contact dermatitis. Methods: Forty mice were randomly divided into 4 groups: blank group, model group, Longdan Xiegan Decoction group and hydrocortisone sodium succinate group, with 10 rats in each group. The dorsal ACD model was prepared by DNCB skin smear method. After the model was successfully replicated, mice in Longdan Xiegan Decoction and hydrocortisone sodium succinate group were given 2 times the equivalent dose of adult gavage for consecutive 21 days. In the blank group, the model group was given the same amount of distilled water . At the end of the experiment, the sensitized skin of mice was used to detect the expression of Foxp3, RORγt and IL-17 mRNA and protein in skin tissues. Results: Compared with the blank control group, the expression of Foxp3 mRNA and protein in the skin of mice in the model control group, the western medicine group and the Longdan Xiegan decoction group were significantly decreased (P <0.01), while the expression of IL-17 and RORγt mRNA and protein The level was significantly higher (P <0.01). Compared with the model control group, the expression of Foxp3 mRNA and protein in the skin of mice in the western medicine group and Longdanxiegan decoction group were significantly increased (P <0.01), while the mRNA and protein expressions of IL-17 and RORγt were significantly decreased (P <0.01). Conclusion: The imbalance of Treg / Th17 cells may be one of the pathogenesis of allergic contact dermatitis. Longdan Xiegan Decoction may regulate the balance of Treg / Th17 cells by acting on the target, Contact dermatitis treatment.
