目的从基因水平探讨过氧化物酶体增殖物激活受体γ2(PPARγ2)基因Pro12Ala多态性对T2DM患者有关指标的影响。方法选取广东省汉族T2DM患者328例,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对PPARγ2基因Pro12Ala多态性的基因型进行检测,并测定相关指标。结果与Pro12Pro组比较,Pro12Ala组T2DM患者TC、LDL-C、瘦素(LP)水平和胰岛素抵抗指数(HOMA-IR)差异有统计学意义(P<0.05)。将所有患者按肥胖组(BMI>25kg/m2,170例)和非肥胖组(BMI≤25kg/m2,158例)分层后,肥胖组TC、TG、LDL-C的水平Pro12Ala组高于Pro12Pro组,差异有统计学意义(P<0.05)。结论 PPARγ2基因Pro12Ala多态性可能影响T2DM患者TC、LDL-C、LP及HOMA-IR。
Objective To investigate the effect of Pro12Ala polymorphism of peroxisome proliferator activated receptor γ2 (PPARγ2) gene on the related indexes of T2DM patients at gene level. Methods A total of 328 T2DM patients were enrolled in this study. Genotypes of PPARγ2 gene Pro12Ala polymorphism were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and the related indexes were determined. Results Compared with Pro12Pro group, the levels of TC, LDL-C, LP and HOMA-IR in T2DM patients in Pro12Ala group were significantly different (P <0.05). The levels of TC, TG and LDL-C in obesity group were higher in Pro12Ala group than those in Pro12Pro group after all patients were stratified by obesity group (BMI> 25kg / m2, 170 cases) and non-obese group (BMI≤25kg / Group, the difference was statistically significant (P <0.05). Conclusion The Pro12Ala polymorphism of PPARγ2 gene may affect TC, LDL-C, LP and HOMA-IR in T2DM patients.
