本文报道人体子宫颈癌HeLa细胞系细胞的超微结构,及其在培养液中加入10,20μg/ml醋酸棉酚,作用24,48h后的变化状态。HeLa细胞在常态生长情况下,具有一般癌细胞的超微结构特点。在醋酸棉酚的作用下,这些细胞间的紧密连接未见发生改变。而在细胞内,线粒体首先发生变化,由肿胀、嵴断裂、空泡化,直至溶解成颗粒状物。棉酚对HeLa细胞的损害作用,具有剂量相关性和作用时间相关性。在棉酚浓度加大和作用时间增长时,可见除线粒体以外的其他细胞器的改变,包括溶酶体大量增多、核膜受损及核内异染色质改变等。扫描电镜下,可见微绒毛断落,细胞表面出现极不正常的泡状突起。本文还对棉酚损伤HeLa细胞的性质及其意义进行了讨论。
This article reports the cell ultrastructure of human cervical cancer HeLa cell line, and its change state after adding 10, 20μg / ml gossypol acetate in culture medium for 24,48h. Under normal growth conditions, HeLa cells have the ultrastructural features of common cancer cells. In the role of gossypol acetate, the tight junctions between these cells have not changed. In the cell, the mitochondria first change, from the swelling, crest fracture, vacuolization, until dissolved into pellets. Gossypol on HeLa cells damage, with dose-dependent and time-dependent. Changes in organelles other than mitochondria, including a large number of lysosomes, impaired nuclear membrane and changes in heterochromatin in the nucleus, were observed when the gossypol concentration was increased and the duration of action increased. Under the scanning electron microscope, the microvilli can be seen to fall off, and abnormally bubbly processes appear on the cell surface. This article also discussed the nature and significance of gossypol injury HeLa cells.
