[目的]探讨白介素-12B(interleukin-12B,IL-12B)基因3′UTR区1188A/C位点多态性与宁夏汉族煤工尘肺(coal worker’s pneumoconiosis,CWP)易感性的关系。[方法]采用聚合酶链反应-限制片段长度多态性(PCR-RFLP)法对120例CWP患者和120例煤尘接触组IL-12B基因1188A/C位点多态性进行分型,对等位基因及基因型分布进行统计学分析。[结果]煤工尘肺组与煤尘接触组在接尘工龄、工种构成、吸烟烟龄差异无统计学意义(P>0.05)。对年龄进行标化后,分析发现IL-12B 1188A/C位点在煤工尘肺组和煤尘接触组中检出三种基因型AA、AC、CC型,这三种基因型在尘肺组和接触组间分布差异无统计学意义(P>0.05);等位基因C在煤工尘肺组和煤尘接触组中所占比例分别为45.8%和36.7%,差异有统计学意义(P<0.05),在接尘工龄、工种和是否吸烟中的分布无差异(P>0.05)。[结论]IL-12B 1188A/C位点C等位基因可能是宁夏汉族煤工尘肺的易感基因之一。
[Objective] To investigate the relationship between 1188A / C polymorphism in 3’UTR region of interleukin-12B (IL-12B) gene and susceptibility to coal worker’s pneumoconiosis (CWP) in Ningxia. [Methods] Polymorphisms of IL-12B gene 1188A / C polymorphism in 120 CWP patients and 120 coal dust exposure groups were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) Allele and genotype distribution for statistical analysis. [Results] There was no significant difference in the service life, composition of work types and smoking age among coal workers’ pneumoconiosis and coal dust exposure groups (P> 0.05). After the standardization of age, we found three genotypes AA, AC and CC were detected in IL-12B 1188A / C locus in coal worker’s pneumoconiosis group and dust contact group. The three genotypes in pneumoconiosis group and There was no significant difference in the distribution among the contact groups (P> 0.05). The allele C was 45.8% and 36.7% respectively in the coal-mining pneumoconiosis group and the coal dust exposure group, with significant difference (P <0.05 ), There was no difference in the occupational age, type of work and whether the smoking was distributed (P> 0.05). [Conclusion] The C allele of IL-12B 1188A / C locus may be one of the susceptible genes of coal worker’s pneumoconiosis in Ningxia Han nationality.
