目的 了解人转铁蛋白 (Tf)与甲氨喋呤 (MTX)结合物 (Tf- MTX)对人肝细胞和人肝癌细胞体外细胞毒的特点。 方法 采用氧化葡聚糖 (Dex) T- 40作为中介体 ,联结 Tf与 MTX制备人转铁蛋白 -甲氨喋呤结合物(Tf- MTX) ,用四甲偶氮唑蓝法比较结合物对人肝细胞 L - 0 2及人肝癌细胞 Bel 740 4的体外杀伤强度。 结果 体外细胞毒试验显示 ,Tf- MTX对人肝细胞 L - 0 2作用 48h的 IC5 0 是对人肝癌细胞 Bel 740 4的 3.36倍 ,而 MTX对上述细胞作用 1h的 IC5 0 是 Tf- MTX的 3.2 2倍。 结论 Tf- MTX对肿瘤细胞 Bel 740 4的杀伤作用有选择性
Objective To investigate the in vitro cytotoxicity of human transferrin (Tf) and methotrexate (MTX) conjugate (Tf-MTX) on human hepatocytes and human hepatoma cells. Methods Dextrans dextran (Tex) 40 was used as the mediator, and Tf and MTX were combined to prepare human transferrin-methotrexate conjugate (Tf-MTX). The conjugates were compared using tetrazolium blue method. In vitro killing strength of human hepatocytes L - 02 and human hepatoma cells Bel 740 4 . RESULTS: In vitro cytotoxicity assay showed that the IC50 of Tf-MTX on human hepatocyte L-02 for 48 h was 3.36 times that of human hepatoma cell Bel 740 4, and the IC50 that MTX exerted on the above cell for 1 h was Tf-MTX. 3.2 2 times. Conclusion Tf-MTX is selective for the killing of tumor cells Bel 740 4
