作者用引物Y3、Y4通过PCR技术对微量陈旧的组织检材进行了扩增,扩增的靶序列位于Y染色体DNA特异3.4Kb重复序列中,产物为446pb。检材是新鲜组织、甲醛固定组织、石蜡包埋组织和HE染色切片组织。检材的保存时间从凡小时到五年。结果表明:新鲜组织,10%中性缓冲甲醛固定一年以下的组织和石蜡包埋3年以下的组织均可做为PCR性别鉴定的良好检材。
The authors used primers Y3 and Y4 to amplify the microscopic old tissue samples by PCR. The amplified target sequence was located on the Y chromosome DNA-specific 3.4kb repeat sequence with a product of 446pb. Samples were fresh tissue, formaldehyde fixation, paraffin-embedded tissue and HE-stained sections. Specimen storage time from hours to five years. The results showed that fresh tissues, tissues fixed under 10% neutral buffered formaldehyde for less than one year and tissues embedded under paraffin for 3 years could be used as good samples for gender identification of PCR.
