目的探讨在不同分化肺鳞癌中基因 p53、p2 1、cerbB - 2、PCNA与DNA含量 (DI值 )的关系。方法采用SP方法检测p53、p2 1、cerbB - 2及PCNA的表达 ;通过Feulgen染色、计算机病理图像分析技术进行DNA含量测定及倍体分析。结果 1 2例高分化肺鳞癌中 ,p53、rasp2 1、cerbB - 0、PCNA阳性率分别为 41 .7%、41 .7%、58.3 %及 66.7% ,DI值均值为 1 .47;1 2例中分化肺鳞癌 p53、rasp2 1、cerbB - 2、PCNA阳性率分别为 58.3 %、66 .7%、66 .7%及83 .3 % ,DI值均值为 1 .74;1 0例低分化鳞癌中 ,p53、rasp2 1、cerbB - 2、PCNA阳性率分别为60 .0 %、60 .0 %、70 .0 %及 90 .0 % ,DI值均值为 2 .1 5。结论 p53、p2 1、cerbB - 2、PCNA阳性率及DNA含量 (DI值)与肺鳞癌的分化程度关系密切 ,其中PCNA阳性率及A×B值、DNA含量 (DI值)与肺鳞癌的分化程度呈正相关 ,P2 1的阳性率与肺鳞癌化程度呈负相关 ,以上多因素综合性性分析 ,对肺鳞癌患者的进一步诊治和预后评估有积极意义
Objective To investigate the relationship between gene p53, p21, cerbB-2, PCNA and DNA content (DI value) in different differentiated lung squamous cell carcinomas. Methods The expression of p53, p21, cerbB-2 and PCNA were detected by SP method. DNA content and ploidy analysis were performed by Feulgen staining and computer pathological image analysis. Results The positive rates of p53, rasp2 1, cerbB - 0 and PCNA were 41.7%, 41.7%, 58.3% and 66.7% in 12 cases of well-differentiated lung squamous cell carcinoma, respectively. The mean value of DI was 1.47; 1 The positive rates of p53, rasp2 1, cerbB-2, and PCNA were 58.3%, 66.7%, 66.7%, and 83.3% respectively in 2 cases of differentiated lung squamous cell carcinoma. The mean DI value was 1.74; 10 cases. In poorly differentiated squamous cell carcinoma, the positive rates of p53, rasp2 1, cerbB-2, and PCNA were 60.0%, 60.0%, 70.0%, and 90.0 %, respectively, and the mean DI value was 2.15%. Conclusions The positive rates of p53, p21, cerbB-2, PCNA and DNA content (DI value) are closely related to the degree of differentiation of squamous cell carcinoma of the lung, among which PCNA positive rate, A×B value, DNA content (DI value) and lung squamous cell carcinoma The degree of differentiation was positively correlated. The positive rate of P21 was negatively correlated with the degree of lung squamous cell carcinoma. Comprehensive analysis of multiple factors above was positive for further diagnosis and treatment of lung squamous cell carcinoma and prognosis evaluation.
