目的与方法 本文用插线法制作局灶性脑缺血 /再灌损伤模型 ,利用激光共聚焦扫描显微镜和fluo 3或DCF DA与PI荧光探剂双标技术检测了活体脑片细胞内脂质过氧化物、Ca2 + 含量及细胞的坏死程度。同时观察巯基供体NAC、S 8744对脑缺血 /再灌损伤的保护作用。结果 ①缺血 /再灌时脑片细胞内脂质过氧化物、Ca2 + 含量增加 ,同时脑片细胞发生坏死 ;②巯基供体NAC、S 8744均可降低缺血 /再灌损伤引起的脑片细胞内脂质过氧化物、Ca2 + 的含量 ,减轻细胞坏死程度。结论 巯基供体NAC、S 8744可拮抗局灶性脑缺血 /再灌损伤
PURPOSE AND METHODS In this study, a focal cerebral ischemia / reperfusion injury model was made by patch-line method. The intracellular lipids in living brain slices were detected by laser scanning confocal microscope, double-labeling with fluo 3 or DCF DA and PI fluorescent probe, Peroxide, Ca2 + content and cell necrosis. Simultaneously observe the protective effect of sulfhydryl donor NAC, S 8744 on cerebral ischemia / reperfusion injury. Results ①In the process of ischemia / reperfusion, the contents of lipid peroxides and Ca2 + in the brain slices increased, and the necrotic cells in the brain slices were also found. ② The NAC and S 8744 groups could reduce the brain damage caused by ischemia / reperfusion injury Intracellular lipid peroxides, Ca2 + content, reduce the degree of cell necrosis. Conclusion Sulfhydryl donor NAC, S 8744 can antagonize focal cerebral ischemia / reperfusion injury
