目的:为考察氧化白藜芦醇在大鼠体内的药动学过程,建立液质联用测定方法。方法:采用液液萃取法对大鼠血浆样品进行预处理,建立以卡马西平为内标,色谱柱为Alltima HPLC C18column(100 mm×2.1 mm,3μm),流速为0.4 mL·min-1的LC-MS/MS检测方法。结果:该方法可以用于大鼠给药氧化白藜芦醇后的血药浓度。结论:该方法耗时短(每个样品仅用3 min),回收率高,专属性强,且准确度和精密度均可满足体内药物分析的要求。
Objective: To investigate the pharmacokinetics of resveratrol in rats and to establish a method for the determination of serum and substance in rats. Methods: The plasma samples of rats were pretreated by liquid - liquid extraction. Carbamazepine was used as internal standard. The column was Alltima HPLC C18column (100 mm × 2.1 mm, 3 μm) at a flow rate of 0.4 mL · min -1 LC-MS / MS detection method. Results: This method can be used for the blood concentration of rats after administration of resveratrol. Conclusion: The method is short time-consuming (only 3 min for each sample), high recovery rate, high specificity, and accuracy and precision can meet the requirements of in vivo drug analysis.
