目的:通过研究干扰素/维甲酸联合应用诱导细胞凋亡相关基因19(GR IM-19)及其靶基因产物转录信号子与激活子3(STAT3)在子宫颈癌组织中的表达情况,探讨GR IM-19及其靶基因产物在子宫颈癌发病中的作用。方法:采用W estern B lotting和RT-PCR方法检测40例子宫颈癌组织和30例正常子宫颈组织中GR IM-19与STAT3及下游基因的表达情况。结果:在子宫颈癌组织中STAT3 mRNA与蛋白均高表达,STAT3下游基因Cyc linB1和Bc l-2L1 mRNA高表达,而GR IM-19mRNA及蛋白表达在癌组织中均明显低于正常组织(P<0.05)。结论:GR IM-19与STAT3在子宫颈癌组织中表达呈负相关,GR IM-19在子宫颈癌组织中的低表达可能与子宫颈癌的发生与发展密切相关。
OBJECTIVE: To investigate the expression of GR IM-19 and its transcriptional product signal transducer and activator of transcription 3 (STAT3) in cervical cancer by interferon / retinoic acid combination therapy The role of GR IM-19 and its target gene products in the pathogenesis of cervical cancer. Methods: The expression of GR IM-19, STAT3 and downstream genes in 40 cases of cervical cancer and 30 cases of normal cervical tissues were detected by Western B lotting and RT-PCR. Results: The expression of STAT3 mRNA and protein in cervical cancer tissues was high, and the expression of CyclinB1 and Bcl-2L1 mRNA in STAT3 was high, while the expression of GR IM-19 mRNA and protein in cancer tissues were significantly lower than that in normal tissues <0.05). Conclusion: The expression of GR IM-19 and STAT3 in cervical cancer tissues is negatively correlated. The low expression of GR IM-19 in cervical cancer tissues may be closely related to the occurrence and development of cervical cancer.
