目的:研究枸杞多糖对小鼠成纤维细胞衰老的延缓作用。方法:从新生昆明小鼠躯干皮肤体外分离培养成纤维细胞,用枸杞多糖处理,分为正常对照组和高、中、低浓度枸杞多糖组,观察细胞生长曲线,采用四甲基偶氮唑蓝(MTT)法检测细胞增殖情况。结果:中、高浓度枸杞多糖可以在传代后6~10天显著增加活细胞数量;MTT实验显示,中、高浓度枸杞多糖组与空白对照组相比,成纤维细胞的相对存活率显著升高,差异具有统计学意义(P<0.05)。结论:枸杞多糖对成纤维细胞的衰老具有拮抗作用,能延迟细胞进入衰老和凋亡状态。
Objective: To study the delaying effect of Lycium barbarum polysaccharide on mouse fibroblast senescence. Methods: Fibroblasts were isolated and cultured from trunk skin of neonatal Kunming mice. The cells were treated with LBP and divided into normal control group and LBP groups. The cell growth curve was observed. (MTT) method to detect cell proliferation. Results: Medium and high concentrations of LBP can significantly increase the number of viable cells 6 to 10 days after passage. The MTT assay showed that the relative survival rate of medium and high LBP groups was significantly higher than that of the blank control group , The difference was statistically significant (P <0.05). CONCLUSION: Lycium barbarum polysaccharides can antagonize the senescence of fibroblasts and delay the cells from entering the senescence and apoptosis.
