目的 研究大鼠服用云南白药后 ,血小板在ADP及花生四烯酸诱导下聚集率的变化 ,以及血小板糖蛋白CD6 1及CD6 2P在静息及ADP诱导条件下在膜表面表达的变化。方法 大鼠ig给药 3d后 ,取血 ,用血小板聚集仪测定血小板的聚集率以及用流式细胞仪测定CD6 1和CD6 2P的表达。结果 无论在ADP或花生四烯酸诱导下 ,云南白药组的血小板聚集率都显著高于对照组 (P <0 .0 1) ;在静息条件下 ,CD6 1及CD6 2P在膜表面的表达虽有少量增加 ,但与对照组相比没有显著差异 (P >0 .0 5 ) ,但在ADP刺激条件下 ,给药组CD6 1及CD6 2P表达比对照组都有显著增加 (P <0 .0 1)。结论 在诱导条件下 ,云南白药可以促进血小板聚集及血小板膜上CD6 1及CD6 2P的表达 ,但不会增加静息血小板表面CD4 1及CD6 2P的表达 ,不会形成促血栓倾向。
Objective To study the changes of platelet aggregation rate induced by ADP and arachidonic acid in rats after taking Yunnanbaiyao, and the changes of platelet glycoproteins CD6 1 and CD6 2P expression on membrane surface under resting and ADP induction conditions. Methods Blood was collected 3 days after ig administration in rats. The platelet aggregation rate was determined by platelet aggregation analyzer and the expression of CD6 1 and CD6 2P was determined by flow cytometry. Results The platelet aggregation rate of Yunnanbaiyao group was significantly higher than that of the control group (P < 0.01) under the induction of ADP or arachidonic acid. The expression of CD6 1 and CD6 2P on membrane surface was observed under resting conditions. Although there was a small increase, there was no significant difference compared with the control group (P > 0.05), but under the ADP stimulation conditions, the expression of CD6 1 and CD6 2P in the drug-administered group was significantly higher than that in the control group (P < 0 .0 1). Conclusion Under inducing conditions, Yunnanbaiyao can promote platelet aggregation and CD6 1 and CD6 2P expression on platelet membrane, but it will not increase the expression of CD4 1 and CD6 2P on resting platelet surface, and will not form prothrombotic tendency.
