目的:验证程序性细胞死亡因子4 (programmed cell death 4,PDCD4)在小鼠雄性生殖细胞中的表达.方法:RT-PCR检测PDCD4在不同周龄小鼠睾丸的基因表达情况;Western blot法检测PDCD4在不同周龄小鼠睾丸中蛋白表达水平;免疫荧光法检测PDCD4在雄性生殖嵴以及在不同周龄小鼠睾丸的定位情况.结果:RT-PCR和Western blot结果均显示PDCD4在0周龄小鼠睾丸中开始有表达,并持续表达至成年;免疫荧光显示PDCD4在胚胎期雄性小鼠生殖细胞中也有表达;在出生后0周和1周龄小鼠睾丸中,PDCD4特异表达于精原细胞;在2周龄小鼠睾丸中,PDCD4分布于精原细胞和精母细胞;而在3、4和5周龄小鼠睾丸组织中,PDCD4主要分布于精原细胞、精母细胞与圆形精子细胞.结论:PDCD4分布于胚胎期和出生后各阶段生精细胞,这为进一步研究PDCD4在精子发生中的作用打下基础.“,”Objective:To validate PDCD4 expression in male mouse germ cells.Methods:We verified Pdcd4 expression in testes of different age mice via RT-PCR and Western blot.The localization of PDCD4 in male reproductive ridge and testis in different age groups was detected by immunofiuorescence.Results:RT-PCR and Westen bolt results showed that PDCD4 was expressed in the 0-week-old mouse testes and continued to express till adulthood;Immunofluorescence showed that PDCD4 was expressed in embryonic male germ cells.PDCD4 was specifically expressed in spermatogonia in the testis of 0 week and 1 week old mice after birth;In the 2-week-old mouse testes,PDCD4 was distributed in spermatogonia and spermatocytes;In the 3 weeks,4 weeks and 5 weeks old mice testicular tissue,PDCD4 mainly distributed in spermatogonia,spermatocytes and round spermatids.Conclusion:PDCD4 is distributed in each level of male germ cells,which lay the foundation for further study of the role of PDCD4 in spermatogenesis.
