目的探讨p130蛋白和mRNA在子宫乳头状浆液性癌(UPSC)中的表达及其临床病理意义。方法收集10例Ⅰ期UPSC患者标本作为UPSC组,10例相同分期高级别子宫内膜癌作为病例对照组,另取相同年龄10例正常增生期子宫内膜标本作为正常对照组。采用HE染色、光镜观察和免疫组织化学方法检测p130蛋白表达水平,实时定量RT-PCR方法检测p130mRNA表达水平,结合临床随访资料进行临床病理分析。结果 UPSC组p130蛋白和mRNA表达(分别为0.46±0.01、0.56±0.06)均明显低于正常增生期子宫内膜(分别为0.91±0.04、2.81±0.40,P<0.01)和高级别子宫内膜癌(分别为0.55±0.03、0.64±0.03,P<0.05)。临床病理分析显示,患者均为绝经后女性,症状均为绝经后阴道不规则出血,肿瘤中位大小7.5cm(1.2~14.8cm),病理学特征与卵巢高级别乳头状浆液性癌相似。结论 UPSC中p130蛋白和mRNA表达减少,可能与UPSC患者预后较差有关。
Objective To investigate the expression of p130 protein and mRNA in uterine papillary serous carcinoma (UPSC) and its clinicopathological significance. Methods 10 cases of stage I UPSC were collected as UPSC group and 10 cases of the same stage high grade endometrial carcinoma as the case control group. Another 10 cases of normal proliferative endometrium of the same age were selected as normal control group. The expression of p130 protein was detected by HE staining, light microscopy and immunohistochemistry. The expression of p130 mRNA was detected by real-time quantitative RT-PCR. Clinicopathological analysis was performed according to clinical follow-up data. Results The protein and mRNA expression of p130 in UPSC group (0.46 ± 0.01 and 0.56 ± 0.06, respectively) were significantly lower than that in normal proliferative endometrium (0.91 ± 0.04, 2.81 ± 0.40, P <0.01, respectively) and high grade endometrium Cancer (0.55 ± 0.03, 0.64 ± 0.03 respectively, P <0.05). The clinicopathological analysis showed that all the patients were postmenopausal women. The symptoms were irregular vaginal bleeding after menopause. The median tumor size was 7.5cm (1.2 ~ 14.8cm). The pathological features were similar to the high grade papillary serous carcinoma in ovary. Conclusion The decrease of p130 protein and mRNA in UPSC may be related to the poor prognosis of UPSC.
