目的:探讨5-氨基乙酰丙酸(5-ALA)介导的光动力(ALA-PDT)对人脑胶质瘤细胞U251的治疗作用。方法:实验于2003-01/2004-05在解放军第一军医大学珠江医院全军神经医学研究所完成。将不同浓度的5-ALA加入到细胞培养基中,随之以激光辐照;固定浓度的5-ALA给予不同剂量的激光辐射。MTT法测定细胞存活率。结果:激光能量恒定于25.0J/cm时,ALA-PDT对U251细胞的杀伤力2在较低5-ALA浓度范围内,随着5-ALA的浓度的增高而增强。各组与对照组间相比较,差异均有显著性意义(F=770.12,P=0.0000)。在较高5-ALA浓度范围内,则存在着饱和现象;相同的5-ALA浓度下,细胞生存率随着激光能量的增加而下降。单用5-ALA或激光处理,均不对细胞造成显著杀伤。结论:5-ALA本身对细胞无明显毒性作用,其介导的PDT是很有前途的胶质瘤治疗方法。
Objective: To investigate the therapeutic effect of 5-aminolevulinic acid (5-ALA) -mediated photodynamic therapy (ALA-PDT) on human glioma U251 cells. METHODS: The experiment was performed at the Institute of Neuromedicine, Zhujiang Hospital, First Military Medical University, January 2003 to May 2004. Different concentrations of 5-ALA were added to the cell culture medium followed by laser irradiation; fixed concentrations of 5-ALA were given different doses of laser radiation. Cell viability was measured by MTT assay. Results: The cytotoxicity 2 of ALA-PDT to U251 cells at a constant laser energy of 25.0 J / cm 2 increased with the increasing concentration of 5-ALA in the lower 5-ALA concentration range. The difference between each group and the control group was significant (F = 770.12, P = 0.0000). At higher 5-ALA concentrations, there was saturation; at the same 5-ALA concentration, cell viability decreased with increasing laser energy. Single treatment with 5-ALA or laser did not cause significant killing of the cells. CONCLUSION: 5-ALA itself has no obvious toxic effect on cells, and PDT mediated by it is a promising method for the treatment of glioma.
