目的探讨骨髓转化生长因子-β1(TGF-β1)及臣核细胞转化生长因子(TGF-β1)Ⅲ型受体(TGF-β1RⅢ)变化在特发性血小板减少性紫癜(ITP)发病中的意义。方法收集急性ITP28例(AITP组)、慢性ITP16例患儿(CITP组)和20例相对正常儿童骨髓;采用Percoll密度梯度及免疫磁珠法分离骨髓巨核细胞;ABC-ELISA法检测TGF-β1水平;原位杂交法检测TGF-β1RⅢmRNA表达情况。结果AITP组与CITP组骨髓TGF-β1水平及巨核细胞TGF-β1RⅢmRNA表达均明显高于对照组(P<0.05),且CITP组明显高于AITP组(P<0.05)。结论检测ITP患儿骨髓TGF-β1水平及巨核细胞TGF-β1RⅢ表达情况对于研究ITP发病机制及早期分型有重要参考价值。
Objective To investigate the significance of TGF-β1 (TGF-β1) and transforming growth factor-β1 (TGF-β1RⅢ) in idiopathic thrombocytopenic purpura (ITP) . Methods Bone marrow was collected from 28 patients with acute ITP (AITP group), 16 patients with chronic ITP (CITP group) and 20 normal children. Percoll density gradient and immunomagnetic beads method were used to isolate bone marrow megakaryocytes. The levels of TGF-β1 ; In situ hybridization detection of TGF-β1R Ⅲ mRNA expression. Results The level of TGF-β1 in bone marrow and the expression of TGF-β1RⅢ mRNA in AITP group and CITP group were significantly higher than those in control group (P <0.05), and were significantly higher in CITP group than in AITP group (P <0.05). Conclusion Detection of TGF-β1 in bone marrow of ITP children and the expression of TGF-β1RⅢ in megakaryocytic cells have important reference value for studying the pathogenesis and early typing of ITP.
