目的:研究痹祺胶囊(BQC)对Ⅱ型胶原诱导的关节炎(CIA)模型大鼠滑膜增殖以及对血清肿瘤坏死因子-α(TNF-α)、白介素-18(IL-18)的影响,揭示痹祺胶囊抗类风湿关节炎(RA)可能的作用途径。方法:采用Ⅱ型胶原建立CIA大鼠模型,按足肿胀度随机分为模型组、BQC高剂量组、BQC中剂量组、甲氨蝶呤(MTX)中剂量组,连续灌胃4周。观察比较各组CIA大鼠后肢肿胀度、血清TNF-α、IL-18水平及HE染色镜下大鼠踝关节滑膜病理改变。结果:与模型组比较,3个用药组足肿胀度明显降低(P<0.05),血清TNF-α、IL-18含量亦有明显降低(P<0.05),关节滑膜增殖及炎性表现较模型组减轻,其中BQC中剂量组疗效更为明显。结论:BQC能够抑制CIA大鼠炎性反应,减轻关节滑膜增殖,可能是通过抑制IL-18、TNF-α的表达而发挥作用。
Objective: To investigate the effects of BQC on synovial proliferation and serum levels of TNF-α and IL-18 in type Ⅱ collagen-induced arthritis rats (CIA) , Revealing biqi capsule anti-rheumatoid arthritis (RA) may be the way of action. Methods: CIA rat model was established by type Ⅱ collagen. The rats were randomly divided into model group, BQC high dose group, BQC middle dose group and methotrexate (MTX) medium dose group for 4 weeks. The degree of hindlimb swelling, serum TNF-α and IL-18 levels and pathological changes of ankle joint synovium of rats with CIA were observed and compared. Results: Compared with the model group, the degree of swelling in the three treatment groups decreased significantly (P <0.05) and the levels of TNF-α and IL-18 in the model group decreased significantly (P <0.05) Model group alleviated, including BQC dose group more obvious effect. CONCLUSION: BQC can inhibit the inflammatory response of CIA rats and reduce the synovial proliferation of synovium, which may be through the inhibition of the expression of IL-18 and TNF-α.
