目的 探讨丁酸对人结肠癌细胞尿激酶型纤溶酶原激活剂(u-PA)及其受体(u-PAR)表达的影响。方法 将人传代结肠癌细胞株SW1116接种于不含或含不同浓度丁酸(2、3、4、7、10 mmol/L)的培养基中,培养6、24、48、72 h后,收集细胞用 ELISA法测定胞质中 u-PA和 u-PAR的表达情况。结果 随着培养基中丁酸浓度的增加和培养时间的延长,经丁酸处理的 SW1116 细胞表达的 u-PA和 u -PAR与对照组相比显著减少,当丁酸浓度≥4 mmol/L和培养时间≥48 h时,两者表达不再增加,反而呈逐步下降趋势。结论 丁酸可抑制SW1116细胞u-PA和u-PAR表达。
Objective To investigate the effect of butyrate on the expression of urokinase-type plasminogen activator (u-PA) and its receptor (u-PAR) in human colon cancer cells. Methods Human colorectal cancer cell line SW1116 was inoculated on medium containing or without different concentrations of butyric acid (2,3,4,7,10 mmol / L) for 6, 24, 48, and 72 h and then collected The expression of u-PA and u-PAR in the cytoplasm was measured by ELISA. Results With the increase of butyrate concentration and incubation time in culture medium, u-PA and u -PAR expression in butyrate-treated SW1116 cells were significantly decreased compared with control group, but when butyrate concentration was ≥4 mmol / L When cultured for more than 48 h, the expression of both did not increase, but decreased gradually. Butyric acid inhibited the expression of u-PA and u-PAR in SW1116 cells.
