目的应用秀丽隐杆线虫评估重金属生物毒性效应。方法将L4期秀丽隐杆线虫暴露于低浓度的CdCl2、CrCl3、As2O3、PbCl2和HgCl2溶液及M9缓冲液(对照组)中72 h,评价不同浓度重金属暴露对线虫生存时间和生殖毒性的影响。结果 2.5、10μmol/L的HgCl2和PbCl2、10μmol/L CdCl2、50μmol/L CrCl3染毒线虫72 h后,与对照组相比,线虫平均寿命均缩短,平均寿命及生存曲线差异均有统计学意义(P均<0.05)。L4期线虫分别在2.5、50、100μmol/L上述重金属溶液中暴露72 h,世代时间延长、后代数目减少,与对照组比较差异均有统计学意义(P均<0.01)。在低浓度时,Hg生殖毒性较Pb、Cd、Cr大,As毒性较弱。结论重金属暴露对秀丽隐杆线虫寿命有一定影响,可致子代生殖能力下降。
Aim To assess the effects of heavy metal toxicity on C. elegans. Methods The exposure of C. elegans L4 to low concentrations of CdCl2, CrCl3, As2O3, PbCl2 and HgCl2 solution and M9 buffer (control group) for 72 h was conducted to evaluate the effects of different concentrations of heavy metal exposure on the survival time and reproductive toxicity of C. elegans. Results After treated with 2.5, 10μmol / L HgCl2 and PbCl2, 10μmol / L CdCl2, 50μmol / L CrCl3 for 72 h, the average life span of nematodes was shortened and the mean life span and survival curve were significantly different (P <0.05). L4 nematodes were exposed to 2.5, 50 and 100μmol / L of the above heavy metals for 72h, respectively. The prolonged generations and the number of offspring decreased compared with the control group (all P <0.01). At low concentrations, Hg reproductive toxicity than Pb, Cd, Cr large, As toxicity is weak. Conclusion Heavy metal exposure has some effect on the lifespan of Caenorhabditis elegans, which may lead to the decline of reproductive capacity of offspring.
