目的检测检测高迁移率族蛋白-1(HMGB-1)在甲状腺乳头状癌(PTC)组织中的表达情况,并探讨其临床意义。方法采用实时荧光定量聚合酶链反应(RT-PCR)法检测50例PTC患者肿瘤组织中HMGB-1的表达水平,同时选择30例结节性甲状腺肿和30例正常人甲状腺组织作为对照。分析PTC癌组织中HMGB-1的表达与PTC临床病理参数间的关系。结果 PTC患者肿瘤组织中HMGB-1 mRNA表达相对水平为(1.47±0.32),明显高于结节性甲状腺肿组织(0.74±0.12)和正常甲状腺组织(0.72±0.09)(P<0.05)。PTC肿瘤组织中HMGB-1的表达与不同TNM分期、腺外侵润、远处转移和淋巴结转移有关(P<0.05)。结论 HMGB-1在PTC肿瘤组织中的表达明显升高,且其表达水平与PTC临床病理特征相关,可能用于PTC早期临床诊断以及对PTC肿瘤浸润、转移的评估。
Objective To detect the expression of high mobility group box-1 (HMGB-1) in papillary thyroid carcinoma (PTC) and to investigate its clinical significance. Methods The expression of HMGB-1 in 50 cases of PTC patients was detected by real-time fluorescent quantitative polymerase chain reaction (RT-PCR). Thirty cases of nodular goiter and 30 cases of normal thyroid tissue were selected as control. To analyze the relationship between the expression of HMGB-1 in PTC tissues and clinicopathological parameters of PTC. Results The relative expression of HMGB-1 mRNA in PTC patients was (1.47 ± 0.32), significantly higher than that in nodular goiter (0.74 ± 0.12) and normal thyroid tissue (0.72 ± 0.09) (P <0.05). The expression of HMGB-1 in PTC tumor tissue was correlated with different TNM stages, extrahepatic invasion, distant metastasis and lymph node metastasis (P <0.05). Conclusion The expression of HMGB-1 in PTC tumor tissues is significantly increased, and its expression level is correlated with the clinical and pathological features of PTC. It may be used in the early clinical diagnosis of PTC and the evaluation of the invasion and metastasis of PTC.
