目的 应用流式细胞术(FCM)对64例大肠癌的石蜡包埋组织的DNA指数(DI)、S期细胞百分比(SPF)、细胞增殖指数(PI)进行检测,并且对上述三项指标与大肠癌的组织类型、组织分化程度、侵犯深度、转移等临床病理因素的关系进行探讨。方法 应用流式细胞仪FACS Calibur,激发波长为488nm,DNA染色试剂为PI,检测上述参数,分析采用Modfit LT2.0软件。结果 在癌组织分化低的大肠癌组中,上述三项指标均高于高—中分化腺癌组,且具有显著意义(P<0.05,P<0.01)。粘液腺癌的SPF、PI值均高于单纯腺癌组(P<0.01)。在有淋巴结转移的大肠癌中DI增高(P<0.05)。在有血行转移的大肠癌中DI及SPF、PI增高(P<0.05,P<0.01)。结论 用石蜡包埋组织进行DNA倍体及细胞周期分析的回顾性研究,能反应肿瘤的生物学特性,参数DI、SPF、PI能够为大肠癌细胞群体的增殖活性与病理因素之间相关性的研究提供依据。
Objective To investigate the DNA index (DI), percentage of S-phase cells (SPF), and cell proliferation index (PI) in paraffin-embedded tissues of 64 cases of colorectal cancer by flow cytometry (FCM). The relationship between the histopathologic factors such as histological type, tissue differentiation, invasion depth, and metastasis of colorectal cancer was discussed. Methods The flow cytometer FACS Calibur was used. The excitation wavelength was 488 nm. The DNA staining reagent was PI. The above parameters were detected and analyzed using Modfit LT2.0 software. Results In the low-differentiation colorectal cancer group, the above three indicators were all higher than those in the high-to-mid-differentiated adenocarcinoma group, with significant significance (P<0.05, P<0.01). The SPF and PI values of mucinous adenocarcinoma were higher than those of simple adenocarcinoma (P<0.01). DI increased in colorectal cancer with lymph node metastasis (P<0.05). DI, SPF, and PI increased in patients with hematogenous metastasis (P<0.05, P<0.01). Conclusion The retrospective study of DNA ploidy and cell cycle analysis in paraffin-embedded tissues can reflect the biological characteristics of tumors. The parameters DI, SPF, and PI can be correlated with the proliferative activity and pathological factors of colorectal cancer cell populations. The study provides the basis.
