目的:研究沙枣可食部分乙醇提取物体内外抗肝肿瘤作用。方法:应用MTT法和移植性H22实体荷瘤小鼠,研究不同剂量提取物的体内体外抗肿瘤作用。结果:提取物对体外培养的人肝癌HepG2细胞增殖有显著的抑制作用,在培养前48 h内,抑制效果与浓度有明显的依赖关系,且提取物的浓度越大,抑制效果越好;在急性毒性试验中,当提取物最大剂量为27.4 g/kg时,对实验小鼠未表现出毒性;荷瘤小鼠模型研究表明,提取物对肝癌H22有显著的抑制作用,当剂量为500 mg/kg时对肿瘤的抑制效果最好,抑瘤率达53.37%,与阳性对照无显著差异;所有沙枣组小鼠的胸腺指数和脾脏指数均显著大于阴性对照组,说明提取物对荷瘤小鼠的免疫功能有显著的促进作用;对肿瘤组织的病理学切片观察表明,提取物具有较强的抗肿瘤作用。
Objective: To study the effect of edible parts of Elaeagnus angustifolia ethanol extract on liver tumor in vitro and in vivo. Methods: MTT assay and transplanted H22 solid tumor-bearing mice were used to study the antitumor effects of different doses of extracts in vitro and in vivo. Results: The extract could significantly inhibit the proliferation of HepG2 cells cultured in vitro. The inhibitory effect was significantly dependent on the concentration within 48 h before culture. The greater the extract concentration, the better the inhibitory effect.
