我们对LDH同功酶的分离、纯化以及免疫动物等技术进行了研究。成功地应用QAE-Sephadex A-50作交换剂,从人红细胞中分离纯化出LDH-1抗原,并获得了兔抗人LDH-1抗体。较文献介绍的其他方法为简便,现介绍如下: 一、材料和试剂配制1.人红细胞。2.QAE-Sephadex A-50(Pharmacia进口分装)。3.洗脱A液:20mM/L Tris缓冲液,pH6.3±0.1。Tris 4.8456g,溶于1600ml蒸溜水中,用6N HCl调至pH6.3,加蒸溜水至2000ml。
We studied LDH isoenzyme separation, purification and immunization of animals and other technologies. Successfully using QAE-Sephadex A-50 as a exchange agent, purified from human erythrocytes LDH-1 antigen, and obtained rabbit anti-human LDH-1 antibody. Other methods described in the literature are simple and are presented below: I. Preparation of Materials and Reagents 1. Human erythrocytes. 2. QAE-Sephadex A-50 (Pharmacia Import Packaged). 3. Elution solution A: 20 mM / L Tris buffer, pH 6.3 ± 0.1. Tris 4.8456g, dissolved in 1600ml distilled water, adjusted to pH6.3 with 6N HCl, add distilled water to 2000ml.
