背景:大脑缺血区白细胞与内皮细胞的活化及表面粘附分子的表达可使白细胞聚集浸润,发挥细胞毒作用进而使脑损伤加重。中药川芎嗪具有抗血栓形成,改善微循环的作用。目的:观察川芎嗪对脑缺血鄄再灌注损伤后局灶区白细胞与内皮细胞粘附性变化的影响。设计:随机对照动物实验。单位:第三军医大学大坪医院脑血管病二科,玉溪市第二人民医院精神科。材料:实验于第三军医大学大坪医院脑血管病二科实验室完成。雄性Wistar大鼠32只,体质量250~300g,随机分为3组,8只为正常对照组,12只为脑缺血鄄再灌注组,12只为脑缺血-再灌注加用川芎嗪组。方法:川芎嗪组静脉注射磷酸川芎嗪注射液(15g/kg)。正常对照组及脑缺血组予静脉注射等量生理盐水。通过免疫荧光标记技术和显微超高速录像系统观察脑缺血-再灌注及应用川芎嗪后局灶区白细胞粘附性的变化。主要观察指标:①脑缺血鄄再灌注损伤局灶区微动脉白细胞附壁密度指数及白细胞与内皮细胞之间黏附力。②应用川芎嗪后局灶区白细胞粘附性的变化。结果:实验动物全部进入结果分析。①与对照组比较,脑缺血鄄再灌注损伤组局灶区微动脉白细胞附壁密度指数增大,白细胞与内皮细胞间断裂应力明显下降,粘附力显著增强,差异均有显著性(P<0.01)。②与脑缺血鄄再灌注损伤组比较,川芎嗪治疗组白细胞附壁密度指数及粘附力显著下降,至24h后降至较低水平,断裂应力增高,至24h与正常对照组差异无显著性,并维持在较高水平。结论:川芎嗪可显著减轻脑缺血鄄再灌注损伤后内皮细胞与白细胞的粘附。
BACKGROUND: The activation of leukocytes and endothelial cells and the expression of surface adhesion molecules in the ischemic area of the brain can cause leukocyte aggregation and infiltration, exerting cytotoxic effects and further aggravating brain damage. The traditional Chinese medicine ligustrazine has antithrombotic effects and improves microcirculation. OBJECTIVE: To observe the effect of tetramethylpyrazine on the adhesion of leukocytes and endothelial cells in focal region after cerebral ischemia-reperfusion injury. Design: Randomized controlled animal experiments. Unit: Department of Cerebrovascular Disease, Daping Hospital, Third Military Medical University, Department of Psychiatry, Second People’s Hospital of Yuxi City. MATERIALS: Experiments were performed at the Second Department of Cerebrovascular Disease, Daping Hospital, Third Military Medical University. Thirty-two male Wistar rats weighing 250-300 g were randomly divided into 3 groups: 8 were normal controls, 12 were cerebral ischaemia reperfusion, and 12 were cerebral ischemia-reperfusion plus ligustrazine group. Methods: Tetramethylpyrazine group was intravenously injected with ligustrazine phosphate injection (15g/kg). Normal control group and cerebral ischemia group were intravenously injected with equal volume of normal saline. The changes of leukocyte adhesion in cerebral ischemic-reperfusion and focal areas after application of tetramethylpyrazine were observed by immunofluorescence labeling and micro-ultrasonic video system. MAIN OUTCOME MEASURES: (1) Density index of leukocyte adhesion to microvessels and adhesion between leukocytes and endothelial cells in focal ischemic/reperfusion injury. 2 Application of tetramethylpyrazine in the focal area of leukocyte adhesion changes. Results: All experimental animals entered the results analysis. 1Compared with the control group, the density index of leukocyte adhesion in the focal region of the cerebral ischemia-reperfusion injury group was increased, the fracture stress between leukocyte and endothelial cells was significantly decreased, and the adhesion force was significantly increased, and the differences were significant (P < 0.05). <0.01). 2Compared with cerebral ischemia-reperfusion injury group, the leukocyte adhesion density index and adhesive force in the tetramethylpyrazine treatment group decreased significantly, and decreased to a lower level after 24 hours, and the fracture stress increased. There was no significant difference from the normal control group at 24 hours. Sex, and maintain at a high level. Conclusion: Tetramethylpyrazine can significantly reduce the adhesion of endothelial cells and leukocytes after cerebral ischemia/reperfusion injury.
