研究了不同浓度APM(4~ 6 μmol/L)和不同处理时间 (4~ 6h)对洋葱根尖细胞有丝分裂的影响。结果表明 ,当用 5 .6 μmol/LAPM处理洋葱分生组织 16h后 ,中期细胞有丝分裂指数 (Met.Ⅰ )从 0 .8(对照 )分别提高至 5 .0、5 .2。结果也同时表明 ,当APM浓度超过 4 μmol/L时 ,严重影响细胞有丝分裂。在根尖细胞中 ,多极分裂细胞 (尤其是 3~ 4级 ) ,中期染色体凝集和微核被检测到。用 2DSDS PAGE分析 ,5种分子量处于 2 4~ 90kD、pI在 5 .0~ 7.3的新的蛋白质被检测到 ,而分子量为 2 4kD、4 0kD ,pI为 4 .8、5 .5的蛋白组分消失。从APM处理后蛋白质组分消失来分析 ,这些蛋白质变化可能与APM处理有关。
The effects of different concentrations of APM (4 ~ 6 μmol / L) and different treatment time (4 ~ 6 h) on the mitosis of root tip cells were studied. The results showed that when treated with 5 .6 μmol / L APM for 16 h, the metaphase mitotic index (Met.Ⅰ) increased from 0.8 (control) to 5.0, 2.5, respectively. The results also showed that when the concentration of APM exceeds 4 μmol / L, seriously affect cell mitosis. In root tip cells, multipolar division cells (especially grades 3-4), metaphase chromosome agglutination and micronuclei are detected. Using 2DSDS PAGE, five new proteins with molecular weights between 24 and 90 kD and pI between 5.0 and 7.3 were detected, while those with molecular weights of 24 kD, 40 kD and pI of 4.8, Points disappear. Analysis of the loss of protein components after APM treatment suggests that these protein changes may be related to APM treatment.
