目的探讨热疗联合5-氟尿嘧啶(5-FU)化疗对小鼠MKN45肿瘤模型细胞凋亡的影响。方法建立裸鼠模型,分为空白对照组、5-FU、热疗组和热疗联合5-FU。流式细胞仪测定凋亡率,免疫组化分析技术(ICH)测定瘤组织表皮细胞生长因子受体(EGFR)和凋亡抑制因子Bcl-2蛋白表达变化。结果:热疗联合5-FU组小鼠MKN45细胞凋亡率最明显(P<0.05),EGFR和Bcl-2蛋白降低最显著(P<0.05)。二者表达呈正相关关系(R=0.914)。结论热疗联合5氟尿嘧啶能增加MKN45肿瘤细胞凋亡可能通过下调EGFR和Bcl-2的表达有关。
Objective To investigate the effect of hyperthermia combined with 5-fluorouracil (5-FU) chemotherapy on the apoptosis of mouse MKN45 tumor model. Methods Nude mice model was established and divided into blank control group, 5-FU, hyperthermia group and hyperthermia combined with 5-FU. The apoptosis rate was determined by flow cytometry. The expression of epidermal growth factor receptor (EGFR) and apoptosis inhibitor Bcl-2 protein in tumor tissues were determined by immunohistochemical analysis (ICH). Results: The apoptosis rate of MKN45 cells in hyperthermia combined with 5-FU group was the most significant (P <0.05), and the decrease of EGFR and Bcl-2 protein was the most significant (P <0.05). The two showed a positive correlation (R = 0.914). Conclusion Hyperthermia combined with 5-Fluorouracil can increase the apoptosis of MKN45 tumor cells by down-regulating the expression of EGFR and Bcl-2.
