目的研究人类着色性干皮病基因D(XPD)Asp312Asn、Lys751Gln基因多态性与铅毒性易感性的关系。方法采用随机整群抽样方法抽取326名铅作业工人为研究对象,检测血铅、尿铅、红细胞锌原卟啉(ZPP)和XPDAsp312Asn、Lys751Gln单核苷酸多态性,根据基因分型分为Asp312Asn GG组与GA+AA组、Lys751Gln AA组与AC+CC组,分析铅作业工人不同XPD基因型与铅毒性易感性的关系。结果 Asp312Asn GG与GA+AA组之间、Lys751Gln AA与AC+CC组之间铅作业工人血铅、尿铅、ZPP水平分别比较,差异均无统计学意义(P>0.05)。分别以血铅≥1.90μmol/L、尿铅≥0.34μmol/L、ZPP≥2.91μmol/L为界将研究对象分为观察组(高水平组)与对照组(低水平组),经2检验与Logistic回归分析显示,Asp312Asn GG与GA+AA组之间、Lys751Gln AA与AC+CC组之间工人高血铅、高尿铅、高ZPP发生率比较,差异均无统计学意义(P>0.05)。结论 XPD Asp312Asn和Lys751Gln基因多态性可能与铅毒性易感性不存在关联。
Objective To study the relationship between polymorphisms of Asp312Asn and Lys751Gln gene and susceptibility to lead toxicity in human dermatophyte dry skin disease gene D (XPD). Methods A total of 326 lead workers were enrolled in this study. Blood lead, urinary lead, zinc oxide protoporphyrin (ZPP) and XPDAsp312Asn, Lys751Gln single nucleotide polymorphisms were detected by genotyping Asp312Asn GG group, GA + AA group, Lys751Gln AA group and AC + CC group. The relationship between different XPD genotypes and susceptibility to lead toxicity in lead workers was analyzed. Results There were no significant differences in blood lead, urinary lead, and ZPP levels between lead-exposed workers and lead-exposed workers between Asp312AsnGG and GA + AA groups (P> 0.05). The subjects were divided into the observation group (high level group) and the control group (low level group) by blood lead≥1.90μmol / L, urinary lead≥0.34μmol / L and ZPP≥2.91μmol / Tests and logistic regression analysis showed that there was no significant difference in the incidence of high blood lead, high urine lead and high ZPP between Asp312Asn GG and GA + AA group, Lys751Gln AA and AC + CC group (P> 0.05). Conclusion The polymorphisms of XPD Asp312Asn and Lys751Gln may not be related to the susceptibility to lead toxicity.
