目的观察阻断磷脂酶C(PLC)-γ1信号通路对卵巢癌细胞凋亡的影响。方法用化学阻断剂U73122处理卵巢癌细胞,光镜观察细胞形态学改变,MTT法评价细胞生长抑制水平,流式细胞仪分析细胞周期变化及细胞凋亡率。结果磷脂酶C信号通路被阻断后,卵巢癌细胞出现明显的细胞凋亡形态学改变,细胞存活率显著下降,流式细胞仪检测到凋亡细胞,与浓度及时间呈一定关系。结论阻断磷脂酶C-γ1能启动卵巢癌细胞的凋亡。
Objective To observe the effect of blocking phospholipase C (PLC) -γ1 signaling pathway on the apoptosis of ovarian cancer cells. Methods The ovarian cancer cells were treated with the chemical inhibitor U73122, the morphological changes of the cells were observed with light microscope, the cell growth inhibition was evaluated by MTT assay, the cell cycle changes and the apoptosis rate were analyzed by flow cytometry. Results After the phospholipase C signal pathway was blocked, the morphological changes of ovarian cancer cells were observed and the cell viability was significantly decreased. The apoptotic cells were detected by flow cytometry, which showed a certain relationship with the concentration and time. Conclusion Blocking phospholipase C-γ1 can induce apoptosis of ovarian cancer cells.
