目的探讨艾滋病(AIDS)患者淋巴结中CD4+CD25+调节性T细胞及增殖细胞核抗原(PCNA)的表达、分布及其之间的关系。方法对35例AIDS患者的淋巴结进行组织学观察和病理分期,采用免疫组织化学法对淋巴结组织中的PCNA及CD4+CD25+调节性T细胞特异性标记物Foxp3进行检测。结果35例艾滋病患者的淋巴结中,1~4期分别有5、4、14和12例。所有淋巴结组织中均检测出Foxp3及PCNA的表达;在各期淋巴结病变中,Foxp3及PCNA的表达模式相似,在1、2期淋巴结病变中,阳性细胞数量较多,主要分布于滤泡间区和付皮质区;3、4期随着淋巴细胞的数量衰竭,其阳性细胞数量减少更显著。Foxp3与PCNA的表达有显著的正相关。结论艾滋病患者淋巴结中的CD4+CD25+调节性T细胞,随着淋巴结病变的进展和细胞增殖活性的变化,其数量逐渐降低或耗竭。
Objective To investigate the expression and distribution of CD4 + CD25 + regulatory T cells and proliferating cell nuclear antigen (PCNA) in lymph nodes of AIDS patients. Methods The lymph nodes of 35 AIDS patients were observed histologically and staged. Immunohistochemical staining was used to detect PCNA and Foxp3, a marker of CD4 + CD25 + regulatory T cells in lymph nodes. Results Among the 35 cases of AIDS patients, there were 5, 4, 14 and 12 cases in stage 1 to stage 4, respectively. The expression of Foxp3 and PCNA were detected in all the lymph node tissues. The expression patterns of Foxp3 and PCNA were similar in all stages of lymphadenopathy. The number of positive cells in stage 1 and 2 was mainly in the interstitial area And pay cortical area; 3, 4 with the decline in the number of lymphocytes, the number of positive cells decreased significantly more. There was a significant positive correlation between Foxp3 and PCNA expression. Conclusions CD4 + CD25 + regulatory T cells in lymph nodes of AIDS patients gradually decrease or disappear with the progression of lymph node lesions and cell proliferation activity.
