为观察高糖、晚期糖化终产物(AGEs)对体外培养的小鼠心脏微血管内皮细胞血管细胞粘附分子 1(VCAM 1)表达的影响 ,分离、培养小鼠心脏微血管内皮细胞 ,以葡萄糖、AGEs单独及联合刺激后流式细胞仪测定内皮细胞VCAM 1的表达 ,放免法测定培养上清中TNF α水平。结果发现 ,高糖、AGEs刺激微血管内皮细胞后VCAM 1表达增加 ,TNFα水平升高 ;联合刺激组VCAM 1表达、TNF α水平更高。提示高糖、AGEs能刺激小鼠心脏微血管内皮细胞VCAM 1表达增加 ,二者具有协同作用 ;TNF α水平的升高与VCAM 1表达增加相一致 ,可能是VCAM 1表达增加的原因之一。
To observe the effects of high glucose and advanced glycation end products (AGEs) on the expression of vascular cell adhesion molecule 1 (VCAM 1) in cultured mouse cardiac microvascular endothelial cells, the mouse cardiac microvascular endothelial cells were isolated and cultured with glucose, AGEs The expression of VCAM-1 in endothelial cells was measured by flow cytometry (FCM) alone and in combination. The level of TNF-alpha in culture supernatant was determined by radioimmunoassay. The results showed that the expression of VCAM-1 increased and the level of TNFα increased after hyperglycemia and AGEs stimulated microvascular endothelial cells. The expression of VCAM-1 and TNF-α in co-stimulated group were higher. These results suggest that high glucose and AGEs can stimulate the expression of VCAM-1 in mouse cardiac microvascular endothelial cells, which has a synergistic effect. The increase of TNF-α is consistent with the increase of VCAM-1 expression, which may be one of the reasons for the increased expression of VCAM-1.
