目的测定广州市2006年Ⅰ型登革病毒流行株的E基因序列并进行分析。方法收集广州市2006年登革热患者急性期血清,用C6/36细胞培养分离登革病毒,RT-PCR法扩增全长E基因,测定序列并绘制系统发生树,进行生物信息学分析。结果59份标本中38份病毒分离培养阳性,获得广州市2006年Ⅰ型登革病毒流行株GZ2006/1707的E基因序列,其同源性与东南亚缅甸、泰国、柬埔寨等地的流行株接近,但与广州市2002年Ⅰ型登革病毒流行株GZ2002/281较远。结论广州市2006年流行的登革病毒属输入性,但与2002年流行的登革病毒有不同的输入源。
Objective To determine and analyze the E gene sequence of epidemic strain of dengue type Ⅰ in Guangzhou in 2006. Methods The dengue virus was isolated from the patients with dengue fever in Guangzhou in 2006. The full length E gene was amplified by RT - PCR, sequenced and phylogenetic tree was constructed for bioinformatics analysis. Results 38 isolates of 59 specimens were positive for isolation and culture, and the E gene sequence of GZ2006 / 1707 strain of dengue type Ⅰ was obtained from Guangzhou in 2006. The homology was close to that of epidemic strains in Southeast Asia, Myanmar, Thailand and Cambodia. However, it was farther away from the GZ2002 / 281 strain of Dengue virus type Ⅰ in Guangzhou in 2002. Conclusion The dengue virus that was prevalent in Guangzhou in 2006 was imported, but it had a different input source than the dengue virus in 2002.
