以金线莲幼嫩茎段为外植体进行组培快繁试验,探索适宜成都地区的金线莲组培繁殖技术。研究结果表明:在初代诱导阶段,采用改良MS+6-BA4.0mg/L+NAA0.5 mg/L的培养基配方可以获得2.5的最大增殖倍数;在增殖阶段,通过调节改良MS培养基中激素水平不能对增殖系数起到明显改善作用,而采用改良B5+6-BA 4.0mg/L+NAA0.5mg/L+2g/L活性炭的组合可以达到3.9的最大增殖倍数,并且继代生根一次完成,说明对金线莲继代增殖基本培养基的筛选是影响成都地区金线莲组培苗生产效率的关键因素。此外,在适当光强度和湿度条件下,采用腐殖土进行栽植60d成活率可达到92%。
Tissue culture and propagation experiments were carried out with tender stem segments of A. asiatica to explore tissue culture techniques of A. japonicus in Chengdu. The results showed that the maximum multiplication factor of 2.5 could be obtained by using the medium formula of MS + 6-BA4.0mg / L + NAA0.5mg / L in the initial induction stage.
