目的 :探讨三氧化二砷 (As2 O3)对结肠癌细胞的凋亡诱导作用及其与细胞周期的关系。方法 :采用透射电镜、丫啶橙荧光染色、TdT及流式细胞仪检测药物作用前后细胞在形态学及细胞周期分布等方面的变化。结果 :形态学观察发现As2 O3 诱导的LoVo细胞死亡呈现凋亡特征 ,研究表明随着药物作用时间延长 ,凋亡细胞比例逐渐增加。As2 O3 在低浓度时主要干扰细胞在S期的通过 ,高浓度时则选择性诱导S期细胞凋亡。结论 :As2 O3 可能是通过诱导细胞凋亡而抑制LoVo细胞增殖 ,这种杀伤作用存在周期特异性。对此研究有助于治疗方案的合理设计。
Objective: To explore the apoptosis-inducing effect of arsenic trioxide (As2O3) on colon cancer cells and its relationship with cell cycle. METHODS: The morphological and cell cycle distributions of the cells before and after drug treatment were detected by transmission electron microscopy, acridine orange fluorescence staining, TdT, and flow cytometry. RESULTS: Morphological observations revealed that As2O3-induced LoVo cell death exhibited apoptosis characteristics. Studies have shown that the proportion of apoptotic cells gradually increases with prolonged drug action. As2O3 mainly interferes with the passage of cells in the S phase at low concentrations, and selectively induces apoptosis in the S phase at high concentrations. Conclusion : As2O3 may inhibit LoVo cell proliferation by inducing cell apoptosis. This killing effect is cycle-specific. This study helps to rationalize the treatment plan.
