作者用体重为35~70克的雌性大鼠,静脉接种伯氏鼠疟原虫NK-65株子孢子1~3×10~6个,于3~10、18~28及29~36小时后,取其肝,以胶原酶法制成含肝脏红外期裂殖体(HEX)的肝细胞悬液。在培养前后,均计数悬液中的肝细胞;以台盼蓝排除试验(trypan blue exclusion)测定肝细胞活力;以腹腔接种大鼠,接种后2~3周内检查有无原虫血症,判定悬液中是否有感染性HEX存在。
In the present study, female rats weighing 35-70 g were inoculated intravenously with 1 to 3 × 10 6 spores of Plasmodium berghei NK-65 strain. After 3 to 10, 18 to 28 and 29 to 36 hours, The liver was taken and collagenase was used to prepare a suspension of hepatocytes containing hepatic inflorescence schizonts (HEX). The hepatocytes in the suspension were counted before and after the culture; the viability of hepatocytes was determined by trypan blue exclusion; the rats were inoculated intraperitoneally and the presence of protozoa was checked within 2 to 3 weeks after inoculation, Susceptible to the presence of infectious HEX in suspension.
