目的:建立固体复合酶活力测定方法及采用固体复合酶测定果糖二磷酸钠(FDP)含量的酶反应方法。方法:在固体复合酶作用下,通过测量还原型辅酶Ⅰ(NADH)在340nm处吸收度的变化来测定果糖二磷酸的含量。结果:果糖二磷酸钠在0.1~0.6mg·mL-1范围内线性关系良好(r=0.9997),注射用果糖二磷酸钠平均回收率为99.3%(n=9);果糖二磷酸钠注射液平均回收率为99.0%(n=9)。结论:本法方法简便、准确,专属性强,可用于果糖二磷酸的含量测定。
Objective: To establish a method for the determination of solid complex enzyme activity and enzyme reaction method using solid complex enzyme to determine the content of fructose diphosphate (FDP). Methods: The content of fructose diphosphate was determined by measuring the change of absorbance of reduced coenzyme Ⅰ (NADH) at 340 nm under the action of solid complex enzyme. Results: The linearity of fructose diphosphate was good in the range of 0.1-0.6 mg · mL-1 (r = 0.9997). The average recovery rate of fructose diphosphate injection was 99.3% (n = 9). The fructose sodium diphosphate injection The average recovery was 99.0% (n = 9). Conclusion: The method is simple, accurate and specific and can be used for the determination of fructose diphosphate.
