目的 研究 2 4 5 0MHz(5mW cm2 )微波 (MW)与 3种化学诱变剂对人淋巴细胞DNA损伤的联合作用。方法 采用彗星试验体外实验检测淋巴细胞经微波和化学诱变剂单独及联合作用后 ,不同培养时间 (0h和 2 1h)的DNA损伤。微波与化学诱变剂联合暴露方式有 3种 :微波辐射后化学诱变剂染毒、微波与化学诱变剂同时暴露、化学诱变剂染毒后微波辐射。 3种化学诱变剂是丝裂霉素C(MMC ,DNA交联剂 )、博来霉素 (BLM ,似X线剂 )、甲基甲烷磺酸酯 (MMS ,烷化剂 )。微波辐射和化学诱变剂暴露时间分别是 2h和 3h。结果 微波组培养 0h和 2 1h ,彗星尾长与对照组比较 ,差异无显著性 (P >0 .0 5 )。培养 2 1h后 ,当MMC浓度为 30 .0 0 μmol L时 ,MW +MMC组、MW MMC组和MMC +MW组的尾长分别为 (18.0 0± 5 .96 )、(2 1.79± 11.4 7)、(2 2 .32± 8.10 ) μm ;当MMC浓度为 3.0 0μmol L时 ,尾长分别为 (8.99± 3.75 )、(12 .4 0± 5 .35 )、(14 .0 0± 5 .38) μm ,明显高于相应的MMC组[(9.4 2± 3.34) μm和 (6 .5 0± 2 .89) μm],差异有显著性 (P <0 .0 1或P <0 .0 5 )。MW +BLM组和MW +MMS组的DNA损伤与相应的BLM组和MMS组比较 ,差异无显著性 (P >0 .0 5 )。结论 2 4 5 0MHz(5mW cm2 )微波辐射 2h未观察到明显的人淋巴细胞
Objective To investigate the combined effects of microwave at 2550MHz (5mW cm2) and three kinds of chemical mutagens on human lymphocyte DNA damage. Methods Comet assay was used to detect the DNA damage of lymphocytes at different culture time (0h and 21h) by microwave and chemical mutagens alone and in combination. Microwave and chemical mutagens combined exposure of three ways: after microwave irradiation chemical mutagen, microwave and chemical mutagens simultaneously exposed, chemical mutagens after exposure to microwave radiation. The three chemical mutagens are mitomycin C (MMC, DNA crosslinker), bleomycin (BLM, like X-ray), methyl methanesulfonate (MMS, alkylating agent). Microwave exposure and chemical mutagen exposure time were 2h and 3h. Results There was no significant difference in the tail length of comet tail between 0 h and 21 h after microwave irradiation (P> 0.05). The tail lengths of MW + MMC group, MW MMC group and MMC + MW group were (18.0 ± 5.96), (2 1.79 ± 11.4 7, respectively) when the concentration of MMC was 30.0 μmol L ), (22.38 ± 8.10) μm, respectively. When the MMC concentration was 3.0 μmol L, the tail lengths were (8.99 ± 3.75), (12.4 ± 5.55) and (14.0 ± 5. 38) μm, which was significantly higher than that of the corresponding MMC group [(9.4 2 ± 3.34) μm vs (6.5 ± 2 .89) μm] (P <0.01 or P <0 .0 5). DNA damage in MW + BLM group and MW + MMS group was not significantly different from corresponding BLM group and MMS group (P> 0.05). Conclusion No obvious human lymphocytes were observed at 2 45 0MHz (5mW cm2) for 2h
