采用 ̄3H-TTP掺入法和DNA聚合酶α、γ选择性抑制剂N-乙基马来酰亚胺,研究了不同剂量、不同剂量率γ射线照射对重组大鼠DNA聚合酶β、BALB/c裸鼠肝细胞和SMMC-LTNM肝癌细胞核内DNA聚合酶β活力的影响。结果表明:受照离体肝细胞和肝癌细胞核内DNA聚合酶β活力无显著改变,重组大鼠DNA聚合酶β在受照40Gy时酶活力比未照组降低15%左右(P<0.05);小鼠整体受照2Gy后,上述两种细胞核内DNA聚合酶β活力在24~28小时内略有下降,随之出现一过性增高,呈现双相变化过程。
Using 3H-TTP incorporation method and DNA polymerase α, γ selective inhibitor of N-ethyl maleimide, different doses of different doses of γ-ray irradiation on recombinant rat DNA polymerase β, BALB / C nude mice hepatocytes and SMMC-LTNM liver cancer cells DNA polymerase beta activity. The results showed that there was no significant change in the DNA polymerase β activity in ex vivo hepatocytes and hepatoma cells, and the enzyme activity of DNA polymerase β was reduced by about 15% compared with that of the control group at 40Gy (P <0.05 ). After the mice were treated with 2Gy as a whole, DNA polymerase β activity in both of the above two nuclei decreased slightly within 24-28 hours, accompanied by a transient increase, showing a biphasic change.
