目的探讨缺氧缺血对新生大鼠脑组织半胱氨酸蛋白酶-1(Caspase-1)的影响。方法新生7日龄SD大鼠112只,结扎左侧颈总动脉后,吸入含8%氧气的氧氮混合气体,制成缺氧缺血性脑损伤(HIBD)模型,于HIBD后12、24 h,3、6、14 d取左侧大脑组织制成匀浆,用Western-blot方法测定Caspase-1蛋白表达量,比较不同时间组间蛋白表达的差异。同时光镜下观察脑组织病理变化。结果缺氧缺血新生大鼠Caspase-1蛋白水平在24 h~6 d逐渐增高,并于6 d达到高峰(0.2839±0.0212),与其时间点前各组相比有显著性差异(P均<0.01),14 d组Caspase-1仍保持较高水平,但与6 d组相比无显著性差异(P>0.05)。结论Caspase-1参与HIBD的形成过程。
Objective To investigate the effect of hypoxia-ischemia on caspase-1 in brain of neonatal rats. Methods One hundred and twelve neonatal SD rats (n = 7) were anesthetized with hypoxia-ischemic brain injury (HIBD) model by inhalation of oxygen and nitrogen mixed gas containing 8% oxygen after ligation of the left common carotid artery. After HIBD, The left brain tissue was made homogenate on day 3, day h, day 3, day 6, and day 14. The expression of Caspase-1 protein was determined by Western-blot and the protein expression differences were compared between different time groups. The pathological changes of brain tissue were observed under light microscope. Results The protein level of Caspase-1 in hypoxic-ischemic neonatal rats increased gradually from 24 h to 6 d and reached the peak at 6 d (0.2839 ± 0.0212), which was significantly different from that before each time point (P < 0.01). Caspase-1 remained high at 14 d, but no significant difference compared with 6 d (P> 0.05). Conclusion Caspase-1 is involved in the formation of HIBD.
