目的探讨谷胱甘肽S转移酶1(GSTP1)和超氧化物歧化酶(SOD)在外阴硬化性苔癣和外阴鳞癌中的表达情况,分析两者与疾病发生发展的关系。方法选取中国医科大学附属第一医院1998—2009年接受手术治疗或活检切除的外阴鳞癌患者30例,外阴硬化性苔藓15例。外阴整形术中去除的正常外阴皮肤组织10例。采用免疫组化方法检测GSTP1和SOD在外阴硬化性苔藓及外阴鳞癌中的表达,并收集外阴鳞癌患者的临床病理资料进行分析。结果 GSTP1和SOD均在正常外阴皮肤表皮基底细胞和真皮层散在的细胞中表达。在外阴鳞状细胞癌中GSTP1和SOD表达均增加(P<0.05)。GSTP1和SOD在Ⅰ~Ⅱ期外阴鳞状细胞癌组织中表达明显低于Ⅲ~Ⅳ期(P<0.05),在高分化中表达明显低于中低分化(P<0.05),在有淋巴结转移中明显高于无淋巴结转移。GSTP1和SOD在外阴鳞癌组织中的表达量呈正相关(r2=0.1474,P=0.01)。在外阴硬化性苔癣中GSTP1表达增多,SOD表达减少(P<0.05)。结论 GSTP1和SOD过表达与外阴鳞癌的发生、发展有关。SOD减少与外阴硬化性苔癣的病变有关。
Objective To investigate the expression of glutathione S-transferase 1 (GSTP1) and superoxide dismutase (SOD) in vulvar lichen vulvar and vulvar squamous cell carcinoma and to analyze the relationship between them and the occurrence and development of the disease. Methods 30 cases of vulvar squamous cell carcinoma and 15 cases of vulva vulgaris were selected from the First Affiliated Hospital of China Medical University from 1998 to 2009 for surgical resection or biopsy. Ten cases of normal vulvar skin tissue removed during vulvar plastic surgery. Immunohistochemistry was used to detect the expression of GSTP1 and SOD in vulvar vulvar lichen and vulvar squamous cell carcinoma, and the clinicopathological data of vulvar squamous cell carcinoma were collected for analysis. Results Both GSTP1 and SOD were expressed in normal vulvar skin epidermal basal cells and scattered cells in the dermis. The expression of GSTP1 and SOD increased in vulvar squamous cell carcinoma (P <0.05). The expression of GSTP1 and SOD in stage Ⅰ ~ Ⅱ vulvar squamous cell carcinoma was significantly lower than that in stage Ⅲ ~ Ⅳ (P <0.05), while in well differentiated stage, the expression of GSTP1 and SOD was significantly lower than that in moderately and poorly differentiated (P <0.05) In significantly higher than without lymph node metastasis. The expression of GSTP1 and SOD in vulvar squamous cell carcinoma was positively correlated (r2 = 0.1474, P = 0.01). In vulvar scleroses, GSTP1 expression increased, SOD expression decreased (P <0.05). Conclusion The overexpression of GSTP1 and SOD is related to the occurrence and development of vulvar squamous cell carcinoma. SOD reduction and vulvar lichen sclerosis lesions related.
