为研究特异性解蛇毒和抗炎症蛋白质药物,对6种江西常见蛇种血清sPLA2抑制蛋白进行筛选和分离纯化。利用实验室前期纯化的五步蛇毒sPLA2作为靶酶,以琼脂糖平板法和pH动态监测法联合筛选江西常见蛇种血清sPLA2抑制剂。基于初筛结果和资源量,确定以华游蛇血清为原料,分离纯化sPLA2抑制剂。分离过程包括Source 30Q、超滤、Uno Q柱精纯化等3个步骤,目的组分经SDS-PAGE分离,显示含有2个亚基,相对分子质量为25.4 ku和21.2 ku。结合文献初步推测该蛋白可能为-γPLI。
In order to study the specific anti-snake venom and anti-inflammatory protein drugs, the sPLA2 inhibitory proteins of six species of common snake in Jiangxi were screened and isolated. Using sPLA2, a purified pentaphylla virus from laboratories, as the target enzyme, sPLA2 inhibitors of common snake species in Jiangxi were screened by agarose plate method and pH dynamic monitoring method. Based on the preliminary screening results and the amount of resources, it was confirmed that sPLA2 inhibitor was isolated and purified from Huakou Serum. The separation process consisted of three steps: Source 30Q, ultrafiltration and Uno Q column purification. The target component was separated by SDS-PAGE and showed two subunits with relative molecular mass of 25.4 ku and 21.2 ku. Combined with the literature presumably the protein may be -γPLI.
