目的探讨口腔鳞癌(OSCC)组织中迁移诱导蛋白7(Mig-7)、血管内皮生长因子(VEGF)和血管生成拟态(VM)的表达及相关临床意义。方法选取80例OSCC患者石蜡标本,采用免疫组织化学染色方法检测其Mig-7、VEGF的表达情况,VM用CD34/PAS双重染色技术确定,并结合OSCC患者临床、病理及随访资料进行相关分析。结果在正常口腔黏膜组织中,VM、Mig-7及VEGF的表达率皆为0%,在80例OSCC组织中三者分别为41.25%、53.75%及71.25%,差异有统计学意义(P<0.05)。其中,VM及Mig-7的阳性表达与肿瘤的分化程度、颈部淋巴结转移情况及五年生存率有关(P<0.05),VEGF的阳性表达与肿瘤的颈部淋巴结转移情况及五年生存率有关(P<0.05),低分化组VM及Mig-7的阳性表达率明显高于中分化组及高分化组,有颈部淋巴结转移组VM、Mig-7及VEGF阳性率高于无颈部淋巴结转移组,VM、Mig-7及VEGF的表达与患者的五年生存率有关,阳性表达组的患者五年生存率明显低于阴性表达组。VM阳性组Mig-7及VEGF的阳性表达率明显高于VM阴性组。结论 OSCC组织中存在VM、Mig-7及VEGF的表达,Mig-7及VEGF的表达极有可能协同促进VM的形成,三者与OSCC的侵袭、转移及较差预后密切相关。
Objective To investigate the expression of migration-induced protein 7 (Mig-7), vascular endothelial growth factor (VEGF) and vasculogenic mimicry (VM) in oral squamous cell carcinoma (OSCC) and its clinical significance. Methods The expression of Mig-7 and VEGF in 80 patients with OSCC was detected by immunohistochemical staining. VM was determined by CD34 / PAS double staining and correlated with the clinical, pathological and follow-up data of OSCC. Results The expression rates of VM, Mig-7 and VEGF in normal oral mucosa tissues were all 0%. Among the 80 OSCC tissues, the expression rates of VM, Mig-7 and VEGF were 41.25%, 53.75% and 71.25% respectively, with significant difference (P < 0.05). The positive expression of VM and Mig-7 correlated with tumor differentiation, cervical lymph node metastasis and five-year survival rate (P <0.05). The positive expression of VEGF and cervical lymph node metastasis and the five-year survival rate (P <0.05). The positive rates of VM and Mig-7 in poorly differentiated group were significantly higher than those in moderately differentiated and well differentiated groups. The positive rates of VM, Mig-7 and VEGF in cervical lymph node metastasis group were higher than those in non-cervical The expression of VM, Mig-7 and VEGF in patients with lymph node metastasis was related to the five-year survival rate of patients. The five-year survival rate of patients with positive expression was significantly lower than that of patients with negative expression. The positive expression rate of Mig-7 and VEGF in VM positive group was significantly higher than that in VM negative group. Conclusion The expression of VM, Mig-7 and VEGF are present in OSCC. The expression of Mig-7 and VEGF is most likely to synergistically promote the formation of VM. The three are closely related to the invasion, metastasis and poor prognosis of OSCC.
