目的:采用血清药理学的方法,观察小半夏加茯苓颗粒对肝癌SMMC-7721细胞的抑制作用。方法:设空白对照组、环磷酰胺(CTX)组、原方汤剂组及颗粒剂不同剂量组,分别制备成含药血清与体外培养的肝癌SMMC-7721细胞共孵,倒置显微镜观察各组SMMC-7721细胞生长形态变化以及用MTT法检测其细胞增殖抑制率。结果:与空白对照组比较,CTX组、原方汤剂组以及颗粒不同剂量组均使SMMC-7721细胞生长形态发生明显改变,细胞生长不良,细胞数量减少;MTT实验结果显示CTX组、汤剂组、颗粒不同剂量组对SMMC-7721细胞作用24h、48h、72h后,细胞增殖抑制率逐渐升高(P<0.05)。各时段内各中药组对细胞的抑制率均低于CTX组(P<0.05),原方汤剂组与颗粒大剂量组无显著差异(P>0.05)。结论:小半夏加茯苓颗粒含药血清对体外培养的肝癌SMMC-7721细胞具有明显的抑制作用,且其作用呈现时间-效应关系和剂量-效应关系。
Objective: To observe the inhibitory effect of Xiao Banxia Jiayu Granules on hepatoma SMMC-7721 cells by serum pharmacology. METHODS: Blank control group, cyclophosphamide (CTX) group, original decoction group and different doses of granules were used to prepare drug-containing serum and in vitro cultured SMMC-7721 cells were co-incubated. Inverted microscopes were used to observe each group. Growth morphological changes of SMMC-7721 cells and inhibition of cell proliferation by MTT assay. RESULTS: Compared with the blank control group, the CTX group, the original decoction group, and the different doses of the particles all significantly changed the growth morphology of the SMMC-7721 cells, resulting in poor cell growth and decreased cell numbers; MTT results showed CTX group and decoction. After 24h, 48h and 72h of SMMC-7721 cells treated with different doses of granules, the inhibitory rate of cell proliferation increased gradually (P<0.05). The inhibitory rate of cells in each group was lower than that in CTX group (P<0.05). There was no significant difference between the original group and high-dose group (P>0.05). Conclusion: Xiao Banxia Jiayu granule drug-containing serum has obvious inhibitory effect on the hepatoma SMMC-7721 cells cultured in vitro, and its time-effect relationship and dose-effect relationship are present.
