目的:研究小鼠骨髓间充质干细胞(BMSCs)对同种异体骨髓来源的树突状细胞(DCs)分化、成熟及功能的影响,探讨MSCs发挥免疫调节作用的机制。方法:体外分离培养BALB/c小鼠BMSCs,与C57BL/6小鼠骨髓有核细胞(BMCs)在体外按不同的细胞比例混合培养,诱导DC分化,流式细胞术(FCM)分析DC细胞表型及FITC-Dextran内吞能力,ELISA检测分泌的细胞因子IL-12的水平。结果:当MSC/BMC达到较高的比例(1∶10)时,细胞表面的CD11c、CD14、CD83、CD86、I-Ab的表达均显著下降(P<0.05),FITC-Dextran内吞能力及IL-12分泌水平亦显著降低(P<0.05)。结论:小鼠MSCs在体外能抑制同种异体骨髓来源的DCs的分化、成熟。
Objective: To investigate the effects of mouse bone marrow mesenchymal stem cells (BMSCs) on the differentiation, maturation and function of allogeneic bone marrow-derived dendritic cells (DCs), and to explore the mechanism of MSCs playing an immunomodulatory role. Methods: BMSCs from BALB / c mice were isolated and cultured in vitro. BMSCs from C57BL / 6 mice were cultured in vitro at different cell ratios to induce DC differentiation. Flow cytometry (FCM) Type and FITC-Dextran endocytosis, ELISA to detect the secretion of cytokines IL-12 levels. Results: The expression of CD11c, CD14, CD83, CD86 and I-Ab on the cell surface was significantly decreased (P <0.05) when the MSC / BMC reached a higher proportion (1:10), and the endocytosis capacity of FITC-Dextran IL-12 secretion was also significantly lower (P <0.05). Conclusion: Mouse MSCs can inhibit the differentiation and maturation of allogeneic bone marrow-derived DCs in vitro.
