目的研究新疆丹参乙酸乙酯提取物的化学成分及化合物Ⅳ对ADP诱导血小板聚集的作用。方法采用溶剂萃取、硅胶等色谱技术手段进行分离纯化,根据化学反应和MS、1HNMR、13CNMR和二维核磁共振等方法进行化合物的结构鉴定;用Born比浊法测定化合物Ⅳ对ADP诱导血小板聚集的抑制作用。结果分离鉴定了6个化合物,分别为二十八烷酸-2-对羟基苯基乙酯(Ⅰ)、β-谷甾醇(Ⅱ)、二十六烷酸(Ⅲ)、6,7-去氢罗列酮(Ⅳ)、3-乙酰齐墩果酸(Ⅴ)、horminone(Ⅵ)。生物活性研究表明:化合物Ⅳ对ADP诱导的血小板聚集具有较好的抑制作用。结论化合物Ⅰ为首次从该科该属该种植物中得到,首次报道化合物Ⅳ具有较强的抑制血小板聚集的作用。
Objective To study the chemical constituents of ethyl acetate extract of Salvia miltiorrhiza and the effect of compound Ⅳ on ADP-induced platelet aggregation. Methods The compounds were isolated and purified by solvent extraction, silica gel and other chromatographic techniques. The structures of compounds were identified by chemical reaction and MS, 1HNMR, 13CNMR and two-dimensional nuclear magnetic resonance. Born turbidimetry was used to determine the effect of compound Ⅳ on ADP-induced platelet aggregation Inhibition. Results Six compounds were isolated and identified as 2-p-hydroxyphenylethyl octacosanoate (Ⅰ), β-sitosterol (Ⅱ), hexacosanoic acid (Ⅳ), oleanolic acid (Ⅴ), horminone (Ⅵ). Biological activity studies show that: compound Ⅳ on ADP-induced platelet aggregation has a good inhibitory effect. Conclusion Compound Ⅰ was first obtained from this genus of this genus, and reported for the first time that compound Ⅳ has a strong inhibitory effect on platelet aggregation.
