目的:探讨肿瘤坏死因子B(TNFB)过度表达时对细胞凋亡、c-JUN、p-JUN与p-JNK表达的影响。方法:采用线栓法制作大鼠大脑中动脉闭塞模型,用TNFB、TUNEL、c-JUN、p-JUN与p-JNK免疫组化染色观察SD大鼠与转小鼠TNFB基因大鼠缺血1h再灌注24h时的细胞凋亡、c-JUN、p-JUN与p-JNK的表达。结果:与SD大鼠相比,转小鼠TNFB基因大鼠缺血1h再灌注24h后,脑组织凋亡细胞显著增多,c-JUN、p-JUN与p-JNK过度表达显著,以上指标变化均显著高于SD大鼠。结论:TNFB的过度表达加剧缺血性脑损伤,其机制可能为c-JUN、p-JUN与p-JNK的过度表达。
Objective: To investigate the effect of tumor necrosis factor B (TNFB) overexpression on the apoptosis, the expression of c-JUN, p-JUN and p-JNK. Methods: The model of middle cerebral artery occlusion was established by thread occlusion. TNFB, TUNEL, c-JUN, p-JUN and p-JNK immunohistochemical staining were used to observe the expression of TNF-α 24h after reperfusion, the expression of c-JUN, p-JUN and p-JNK. Results: Compared with SD rats, the apoptotic cells in the brain of TNFB transgenic mice were significantly increased at 1 h and 24 h after ischemia, and c-JUN, p-JUN and p-JNK were over-expressed Were significantly higher than SD rats. CONCLUSION: Overexpression of TNFB aggravates ischemic brain injury and its mechanism may be overexpression of c-JUN, p-JUN and p-JNK.
