以玉米自交系齐319、郑58、昌7-2、Lx9801和81162的成熟胚为外植体诱导愈伤组织,研究了基因型、外源激素组合、低温处理对愈伤组织诱导的影响。结果表明,基因型对玉米成熟胚愈伤组织诱导的影响显著。在供试的5个基因型中,齐319的愈伤组织诱导率最高,诱导出的愈伤组织质量也较好,为致密、松脆、淡黄色或乳白色,昌7-2不仅愈伤组织诱导率低而且不能产生胚性愈伤组织:在所试验的外源激素组合中,以4,0 mg/L 2,4-D+0.5 mg/L 6-BA对愈伤组织的诱导最为有效;4℃低温预处理3 d对愈伤组织的诱导具有明显的促进作用。经处理,齐319愈伤组织的诱导率由67.5%提高到78.3%。研究确定了愈伤组织诱导的最佳条件,为建立高效的玉米成熟胚再生体系奠定了基础。
The callus was induced by mature embryos of maize selfing lines Qi 319, Zheng 58, Chang 7-2, Lx9801 and 81162. The effects of genotype, exogenous hormones combination and low temperature treatment on callus induction were studied. . The results showed that the genotype had a significant effect on the induction of callus from maize embryo. Among the five genotypes tested, the callus induction rate of Qi 319 was the highest, and the callus induction quality was also good, which was dense, crisp, light yellow or milky white.
