目的 建立超高效液相色谱-质谱法测定小鼠角膜中环孢素的含量.方法 流动相:甲醇-水(90∶10),色谱柱:Hypersill C18柱(100 mm×2.1 mm, 1.9 μm),内标物:环孢素A.电离方式为ESI 正离子模式,用多反应监测模式,环孢素:m/z 1224.9 [ M +Na]+.结果 角膜匀浆液中环孢素的线性范围为10~200 ng· mL-1(R=0.9986),最低检测质量浓度为3.3 ng· mL-1.鼠角膜中环孢素的平均提取回收率为80%(n=6),RSD<12%.日内精密度RSD均小于10.12%,日间精密度RSD均小于11.75%.结论 本方法灵敏度高、专属性强及重复性好,可快速准确地测定小鼠角膜中环孢素的质量浓度.“,”Objective To establish a ultra performance liquid chroma-tography-mass spectrometry to determine the content of cyclosporine in mice.Methods Mobile phase was methanol -water (90∶10), chroma-tographic column was: Hypersill C18column (100 mm×2.1 mm, 1.9 μm) and internal standard substance : Cyclosporine A, ion mode of ESI +was applied to determine cyclosporine and multi -response monitoring mode was used (MRM, cyclosporine m/z: 1224.9 ).Results Working solution over the concentration range of 10 ~200 ng · mL-1 (R=0.9986) worked well for the determination of cyclosporine in mice cornea homogenate.The lowest concentration was 3.3 ng· mL-1.The average extraction recovery rate of cyclosporine in mice cornea reached almost 80%(n=6), and RSD was less than 12%.The intra-precision was less than 10.12%and the inter -precision was less than 11.75%. Conclusion The established method was sensitivity , specific, repea-table for cyclosporine determination in mice cornea .
