目的通过结晶硫化镍在支气管上皮细胞系(16 HBE)中磷酸化氨基末端蛋白激酶(P-JNK)和P38的表达,探讨镍致癌的分子机制和中草药对职业性肿瘤的防治作用。方法用SDS聚丙烯酰胺凝胶电泳和蛋白免疫印迹杂交(Western blot)法检测16 HBE细胞中P-JNK和P38的表达。结果硫化镍能明显激活P-JNK和P38的表达,硫化镍浓度越高,磷酸化P-JNK和P38的表达值越高,而且存在剂量-效应关系,染镍组与对照组比较,差异有统计学意义(P<0.05、P<0.01)。结论结晶型硫化镍激活16 HBE细胞中P-JNK和P38的表达,可能是镍致癌的分子机制之一,扶正解毒含药血清对职业性肿瘤有一定的防治作用。
Objective To investigate the molecular mechanism of nickel carcinogenesis and the preventive and therapeutic effects of Chinese herbal medicine on occupational tumors by the expression of phospho-N-terminal kinase (P-JNK) and P38 in bronchial epithelial cell line (16 HBE). Methods The expressions of P-JNK and P38 in 16 HBE cells were detected by SDS polyacrylamide gel electrophoresis and Western blot. Results Nickel sulphide significantly activated the expressions of P-JNK and P38. The higher the nickel sulphide concentration, the higher the expression of phosphorylated P-JNK and P38, and there was a dose-effect relationship. Compared with the control group, the difference was Statistical significance (P <0.05, P <0.01). Conclusion The expression of P-JNK and P38 in 16 HBE cells induced by crystalline nickel sulfide may be one of the molecular mechanisms of nickel carcinogenesis. Fuzheng detoxification serum may have some preventive and therapeutic effects on occupational tumors.
