目的比较研究10种人造矿物纤维(MMMF)对人肺泡上皮细胞(A549细胞)DNA的损伤及修复作用。方法选用日本纤维状物质研究协会提供的10种标准人造矿物纤维(JFM),用单细胞凝胶电泳法,以A549细胞作为靶细胞,测定了细胞DNA链断裂、损伤后修复及DNADNA链间交联。结果10种JFM纤维都可引起不同程度的DNA链断裂、DNADNA链间交联及抑制DNA损伤后的修复功能。综合分析发现:微细玻璃纤维的遗传毒性最强,硅灰石的遗传毒性最低。它们的作用程度均低于温石棉。结论JFM纤维均具有一定程度的体外遗传毒性,但均低于UICC温石棉B。
Objective To compare the DNA damage and repair of human alveolar epithelial cells (A549 cells) with 10 kinds of man-made mineral fibers (MMMF). Methods Ten kinds of standard artificial mineral fibers (JFM) from Japan Fibrous Material Research Association were selected. Using single cell gel electrophoresis and A549 cells as target cells, the cell DNA strand breaks, DNA damage and DNA DNA strand breaks Between the cross-linked. Results 10 kinds of JFM fibers can cause different degrees of DNA strand breaks, cross-links between DNA-DNA strands and inhibit DNA damage repair function. Comprehensive analysis found that: the finest genetic toxicity of glass fiber strongest wollastonite genotoxicity lowest. Their extent of action is lower than chrysotile. Conclusions JFM fibers have a certain degree of genotoxicity in vitro, but both are lower than that of UICC chrysotile B.
