目的探讨大蒜素对体外培养的人宫颈腺癌Hela、鳞癌Cask i细胞半胱氨酸蛋白酶(Caspase)-3、8、9的表达和活性变化对宫颈癌细胞生长的影响及其作用机制。方法2008年1月至2009年1月在河北医科大学附属第二医院通过体外培养的人宫颈腺癌Hela、鳞癌Cask i细胞,MTT法检测大蒜素作用后细胞的生长抑制率,分光光度法和RT-PCR方法分别检测大蒜素对细胞Caspase-3、8、9活性和表达的影响。结果用6.25、12.5、25、50、75、100mg/L的大蒜素处理Hela、Cask i细胞24、48、72h后,随着药物浓度和作用时间的增加,抑制率增高。分光光度法检测大蒜素作用48h后,Hela细胞Caspase-3、8、9活性的OD实验组/OD对照组分别是对照组的2.32、2.26、1.53倍;Cask i细胞是对照组的2.28、1.69、2.08倍(P<0.05)。RT-PCR法检测大蒜素作用12h后,Caspase-3、8、9mRNA基因扩增产物与β-actin基因扩增产物电泳带的吸光度比值明显升高,Hela细胞为对照组的1.92、1.47、1.57倍,Cask i细胞为对照组的1.87、1.55、1.66倍(P<0.05)。结论大蒜素对宫颈癌Hela和Cask i细胞有抑制作用,随时间和剂量增加抑制率上升。
Objective To investigate the effects and mechanisms of allicin on the growth of human cervical adenocarcinoma Hela and squamous cell carcinoma of Cask i cells and the changes of the expression and activity of caspase -3,8,9. Methods From January 2008 to January 2009, the growth inhibition rates of allogeneic cervical adenocarcinoma Hela, squamous cell carcinoma Cask i and MTT were detected in the Second Affiliated Hospital of Hebei Medical University. The inhibitory rate of allogeneic cells was determined by spectrophotometry The effects of allicin on the activity and expression of Caspase-3, 8, 9 were detected by RT-PCR. Results Hela and Cask i cells were treated with allicin at 6.25, 12.5, 25, 50, 75 and 100 mg / L for 24, 48 and 72 hours, respectively. With the increase of drug concentration and time, the inhibitory rate increased. 48 hours after the allicin was detected by spectrophotometry, the activity of Caspase-3, 8 and 9 in Hela cells was 2.32, 2.26 and 1.53 times higher than that of the control group and 2.28 and 1.69 respectively in the control group , 2.08-fold (P <0.05). After the allicin was detected by RT-PCR for 12h, the ratio of absorbance of the amplification products of Caspase-3, 8, 9 mRNA and β-actin gene was significantly increased, Hela cells were 1.92,1.47,1.57 Fold, Cask i cells 1.87,1.55,1.66 times the control group (P <0.05). Conclusion Allicin has an inhibitory effect on cervical cancer Hela and Cask i cells. With the increase of time and dosage, the inhibitory rate increases.
